Abstract This report is a study of the effects of UV exposure on the growth of four strains of non-photosynthetic bacteria Escherichia coli Staphylococcus aureus Bacillus megaturium and Psuedomonas aeruginosa Using a UV Transilluminator each strain was exposed to nine different time intervals of UV radiation at a frequency of 1x 10 Mhz After 48 hours of incubation the kill time was determined by the first interval that produced no visible growth of colonies The kill time for each strain of bacteria are as follows S aureus-10 to 20 minutes B megaturium-5 to 10 minutes Ps aeruginosa- 2 to 5 minutes E coli- 20 to 40 seconds Staphylococcus aureus showed 30 times more resistance to UV radiation than Escherichia coli This resistance might be due to the Gram-Positive cocci shape of S aureus Or aerobic bacteria have adapted metabolic pathways that enable them to repair the Thymine dimers created by UV radiation I recommend research investigating DNA polymerase and other enzymes capable of repairing DNA mutations to determine their effectiveness against Thymine Dimers and how they vary among the different strains of bacteria Introduction Food production companies are constantly researching for better and more cost-efficent ways of pasteurizing food One possible method is Ultra Violet UV Irradiation The same kind of light emitted by the sun and linked to skin cancer UV radiation works by forming kinks in the cells DNA that prevent the cell from reproducing These kinks are caused by bonds that form between adjacent thymine molecules forming a Thymine Dimer To a certain degree humans have natural defenses to protect against Thymine dimers This experiment will determine whether different strains of bacteria are capable of protecting against UV radiation and to what degree The variable for this experiment will be time We will expose the bacteria to UV light at varying time intervals 10 seconds 20s 40s 25
